J Strength Cond Res. 25(4): 973-979, 2011-Australian football requires frequent intermittent sprinting close to peak running speed. However, tests assessing the capability to maintain intermittent peak running speed are not reported in scientific literature. Therefore, our objective is to report the reliability and validity of a novel intermittent peak running speed test. The intermittent peak running speed test required footballers to perform 10 repetitions on 25-second intervals. Each repetition required 15-m jogging, 20-m acceleration to peak speed, 10 m to sustain peak speed, 20-m deceleration, and finally a 15-m jog. Intermittent peak running speed was determined by portable global positioning system. To assess reliability, 26 footballers performed the intermittent peak running speed test on 2 occasions 3-5 days apart. Our results revealed that average peak speed had a coefficient of variation of 2.2% and an intraclass correlation of 0.91. To assess construct validity, average peak speed was compared between elite, sub-elite, and regional footballers. The average peak speed of the elite footballers (28.6 ± 1.7 km-h-1) was higher than that of the sub-elite (27.4 6 1.7 km-h -1) and regional (27 6 1.9 km-h-1) competitors (p < 0.05). Our study revealed that the intermittent peak running speed test possesses acceptable reliability and distinguishes between elite and sub-elite footballers. © 2011 National Strength and Conditioning Association.
Soil moisture, organic carbon, and nitrogen content prediction with hyperspectral data using regression models

- Description: Soil moisture, soil organic carbon, and nitrogen content prediction are considered significant fields of study as they are directly related to plant health and food production. Direct estimation of these soil properties with traditional methods, for example, the oven-drying technique and chemical analysis, is a time and resource-consuming approach and can predict only smaller areas. With the significant development of remote sensing and hyperspectral (HS) imaging technologies, soil moisture, carbon, and nitrogen can be estimated over vast areas. This paper presents a generalized approach to predicting three different essential soil contents using a comprehensive study of various machine learning (ML) models by considering the dimensional reduction in feature spaces. In this study, we have used three popular benchmark HS datasets captured in Germany and Sweden. The efficacy of different ML algorithms is evaluated to predict soil content, and significant improvement is obtained when a specific range of bands is selected. The performance of ML models is further improved by applying principal component analysis (PCA), a dimensional reduction method that works with an unsupervised learning method. The effect of soil temperature on soil moisture prediction is evaluated in this study, and the results show that when the soil temperature is considered with the HS band, the soil moisture prediction accuracy does not improve. However, the combined effect of band selection and feature transformation using PCA significantly enhances the prediction accuracy for soil moisture, carbon, and nitrogen content. This study represents a comprehensive analysis of a wide range of established ML regression models using data preprocessing, effective band selection, and data dimension reduction and attempt to understand which feature combinations provide the best accuracy. The outcomes of several ML models are verified with validation techniques and the best- and worst-case scenarios in terms of soil content are noted. The proposed approach outperforms existing estimation techniques.
Recovering the “individual” for qualitative research: An idiographic approach

- Description: As detailed examination of the experience of the individual, the Self or the I is overtaken in the intellectual climate of qualitative research by an aim to understand human experience on a collective or transferable level, the claim made by qualitative researchers to providing genuine understanding of the “what is it like” characteristics of being human arguably becomes shaky. If the wellspring from which we draw our understanding is limited to understandings that researchers recognize as general, then the unique and deeper characteristics of individual experience may be buried within the aggregate. We contend that any such restricted approach cannot begin by itself to cogently inform a theory of or a theory for examining human experience that is sufficiently sophisticated for qualitative research practice. Consequently, we propose a recovery and inclusion, into qualitative research frameworks, of a strongly idiographic consideration of the “what is it like” characteristics of phenomena, as experienced by the individual person. Recommending thereby a recovery of hermeneutic and phenomenological modes of thought, in this article, we suggest that the central ideas of KELLY’s personal construct psychology involve fertile ground for guiding such a shift in qualitative research. © 2019, Institut für Qualitative Forschung,Internationale Akademie Berlin gGmbH. - Description: MAIT cells are MR1-restricted T cells that are well-known for their anti-microbial properties, but they have recently been associated with different forms of cancer. Several studies have reported activated MAIT cells within the microenvironment of colorectal tumors, but there is conjecture about the nature of their response and whether they are contributing to anti-tumor immunity, or to the progression of the disease. We have reviewed the current state of knowledge about the role of MAIT cells in colorectal cancer, including their likely influence when activated and potential sources of stimulation in the tumor microenvironment. Knock down of TIMP-2 by siRNA and CRISPR/Cas9 mediates diverse cellular reprogramming of metastasis and chemosensitivity in ovarian cancer

- Description: Background: The endogenous tissue inhibitor of metalloproteinase-2 (TIMP-2), through its homeostatic action on certain metalloproteinases, plays a vital role in remodelling extracellular matrix (ECM) to facilitate cancer progression. This study investigated the role of TIMP-2 in an ovarian cancer cell line in which the expression of TIMP-2 was reduced by either siRNA or CRISPR/Cas9. Methods: OVCAR5 cells were transiently and stably transfected with either single or pooled TIMP-2 siRNAs (T2-KD cells) or by CRISPR/Cas9 under the influence of two distinct guide RNAs (gRNA1 and gRNA2 cell lines). The expression of different genes was analysed at the mRNA level by quantitative real time PCR (qRT-PCR) and at the protein level by immunofluorescence (IF) and western blot. Proliferation of cells was investigated by 5-Ethynyl-2′-deoxyuridine (EdU) assay or staining with Ki67. Cell migration/invasion was determined by xCELLigence. Cell growth in vitro was determined by 3D spheroid cultures and in vivo by a mouse xenograft model. Results: Approximately 70–90% knock down of TIMP-2 expression were confirmed in T2-KD, gRNA1 and gRNA2 OVCAR5 ovarian cancer cells at the protein level. T2-KD, gRNA1 and gRNA2 cells exhibited a significant downregulation of MMP-2 expression, but concurrently a significant upregulation in the expression of membrane bound MMP-14 compared to control and parental cells. Enhanced proliferation and invasion were exhibited in all TIMP-2 knocked down cells but differences in sensitivity to paclitaxel (PTX) treatment were observed, with T2-KD cells and gRNA2 cell line being sensitive, while the gRNA1 cell line was resistant to PTX treatment. In addition, significant differences in the growth of gRNA1 and gRNA2 cell lines were observed in in vitro 3D cultures as well as in an in vivo mouse xenograft model. Conclusions: Our results suggest that the inhibition of TIMP-2 by siRNA and CRISPR/Cas-9 modulate the expression of MMP-2 and MMP-14 and reprogram ovarian cancer cells to facilitate proliferation and invasion. Distinct disparities in in vitro chemosensitivity and growth in 3D culture, and differences in tumour burden and invasion to proximal organs in a mouse model imply that selective suppression of TIMP-2 expression by siRNA or CRISPR/Cas-9 alters important aspects of metastasis and chemosensitivity in ovarian cancer. © 2022, The Author(s).

- Description: Catastrophic megafires can increase extinction risks identifying species priorities for management and policy support is critical for preparing and responding to future fires. However, empirical data on population loss and recovery post-fire, especially megafire, are limited and taxonomically biased. These gaps could be bridged if species' morphological, behavioural, ecological and life history traits indicated their fire responses. Using expert elicitation that estimated population changes following the 2019–20 Australian megafires for 142 terrestrial and aquatic animal species (from every vertebrate class, one invertebrate group), we examined whether expert estimates of fire-related mortality, mortality in the year post-fire, and recovery trajectories over 10 years/three generations post-fire, were related to species traits. Expert estimates for fire-related mortality were lower for species that could potentially flee or shelter from fire, and that associated with fire-prone habitats. Post-fire mortality estimates were linked to diet, diet specialisation, home range size, and susceptibility to introduced herbivores that damage or compete for resources. Longer-term population recovery estimates were linked to diet/habitat specialisation, susceptibility to introduced species species with slower life histories and shorter subadult dispersal distances also had lower recovery estimates. Across animal groups, experts estimated that recovery was poorest for species with pre-fire population decline and more threatened conservation status. Sustained management is likely needed to recover species with habitat and diet specialisations, slower life histories, pre-existing declines and threatened conservation statuses. This study shows that traits could help inform management priorities before and after future megafires, but further empirical data on animal fire response is essential.

- Description: The importance of relating with God for Spiritual Well-Being Twenty years of research in spiritual health/well-being culminate in this thesis. My PhD (1998), University of Melbourne, presented a Four Domains Model of Spiritual Health/Well-Being. This formed the theoretical base for several spiritual well-being questionnaires, presented in my EdD (2009), University of Ballarat. The main instrument, Spiritual Health And Life-Orientation Measure (SHALOM), proposed a novel method of assessing quality of relationships which are posited as reflecting spiritual well-being. These relationships are of each person with themselves, others, the environment and/or with a Transcendent Other (God). Instead of just investigating lived experiences, SHALOM asks for two responses to each item. Each respondent’s lived experience is compared with their stated ideals. Studies presented herein show that this double-response method, comparing ideal with actual, provides a statistically stronger measure of quality of relationships than lived experiences alone, thus spiritual well-being. Eighteen publications included in this thesis relate to studies of spiritual well-being in educational, healthcare and general community settings. A total of 52 studies, using SHALOM with 41686 people from 27 countries, have shown that, of the four sets of relationships measured by SHALOM, relating with God explains greatest variance in spiritual well-being, so is therefore most important. An international study with people involved with, as well as some opposed to, spirituality and religious education led to a generic form of SHALOM, in which the theistic words were replaced by ‘Transcendent’, with 19 alternatives provided from which to select preferred Transcendent, or none. This project revealed that relating with God, as Transcendent, provides greatest support for a person’s relating with self and others, in contrast to help provided by non-theistic and non-religious Transcendents and not believing in Transcendents. This research implies that researchers need to investigate relationship with God in any study of spiritual well-being, whether or not it fits their personal worldview.

- Description: Monitoring of structures to identify types of damages that occur under loading is essential in practical applications of civil infrastructure. In this paper, we detect and visualize damage based on several non-destructive testing (NDT) methods. A machine learning (ML) approach based on the Support Vector Machine (SVM) method is developed to prevent misdirection of the event interpretation of what is happening in the material. The objective is to identify cracks in the early stages, to reduce the risk of failure in structures. Theoretical and experimental analyses are derived by computing the performance indicators on the smart aggregate (SA)-based sensor data for concrete and reinforced-concrete (RC) beams. Validity assessment of the proposed indices was addressed through a comparative analysis with traditional SVM. The developed ML algorithms are shown to recognize cracks with a higher accuracy than the traditional SVM. Additionally, we propose different algorithms for microwave- or millimeter-wave imaging of steel plates, composite materials, and metal plates, to identify and visualize cracks. The proposed algorithm for steel plates is based on the gradient magnitude in four directions of an image, and is followed by the edge detection technique. Three algorithms were proposed for each of composite materials and metal plates, and are based on 2D fast Fourier transform (FFT) and hybrid fuzzy c-mean techniques, respectively. The proposed algorithms were able to recognize and visualize the cracking incurred in the structure more efficiently than the traditional techniques. The reported results are expected to be beneficial for NDT-based applications, particularly in civil engineering.

- Description: The air-entry value (AEV) suction, marking the transition between saturated and unsaturated soil mechanics, is arguably the most important parameter interpreted from the soil–water characteristic curve (SWCC); its accurate determination being essential for the prediction of unsaturated soil properties. The AEV is commonly obtained by a subjective and time-consuming graphical construction. This micro-article proposes an objective framework, developed based on a practical mathematical translation technique, for the AEV determination. Explicit equations for the AEV are derived based on eleven well-established SWCC fitting functions, covering a wide range of functional complexities. In addition to its objective nature (providing unique interpretations of the AEV), the proposed framework complements numerical implementations of unsaturated soil constitutive models. © 2021 The Author(s)
Efficient data gathering in 3D linear underwater wireless sensor networks using sink mobility

- Description: Due to the unpleasant and unpredictable underwater environment, designing an energy-efficient routing protocol for underwater wireless sensor networks (UWSNs) demands more accuracy and extra computations. In the proposed scheme, we introduce a mobile sink (MS), i.e., an autonomous underwater vehicle (AUV), and also courier nodes (CNs), to minimize the energy consumption of nodes. MS and CNs stop at specific stops for data gathering; later on, CNs forward the received data to the MS for further transmission. By the mobility of CNs and MS, the overall energy consumption of nodes is minimized. We perform simulations to investigate the performance of the proposed scheme and compare it to preexisting techniques. Simulation results are compared in terms of network lifetime, throughput, path loss, transmission loss and packet drop ratio. The results show that the proposed technique performs better in terms of network lifetime, throughput, path loss and scalability. © 2016 by the authors; licensee MDPI, Basel, Switzerland.

A modified MTS proliferation assay for suspended cells to avoid the interference by hydralazine and β-Mercaptoethanol

- Description: The 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium (MTS) assay is one of the most commonly used tests of cell proliferation. Hydralazine has been reported to interfere with the performance of the MTS assay when used on adherent cells. This study aimed to investigate whether hydralazine interferes with the performance of the MTS assay on suspended cells. THP-1 (a monocytic leukemia cell line) cells were cultured in the presence or absence of hydralazine (0, 10, 50, 100, and 500 μM) for 2 or 24 h. Cell numbers were analyzed using the MTS, trypan blue exclusion, or microscopic assays. A modified version of the standard MTS assay was established by centrifuging the cells and replacing the test medium with fresh culture medium immediately before the addition of the MTS reagent. Culture of THP-1 cells with hydralazine at concentrations of 50, 100, and 500 μM for 2 h increased absorbance (p < 0.001) in the standard MTS assay, whereas both the trypan blue exclusion assay and microscopy suggested no change in cell numbers. Culture of THP-1 cells with 100 and 500 μm hydralazine for 24 h increased absorbance (p < 0.05) in the standard MTS assay; however, trypan blue exclusion and microscopy suggested a decrease in cell numbers. In a cell-free system, hydralazine (100 and 500 μM) increased absorbance in a time- and concentration-dependent manner. The modified MTS assay produced results consistent with trypan blue exclusion and microscopy using THP-1 cells. In addition, the modified MTS assay produced reliable results when K562 and Jurkat cells were incubated with hydralazine or β-mercaptoethanol (βME). - Description: The growth experiences of women-owned Small and Medium Enterprises (SMEs) in the context of a developing economy are examined through the lens of pragmatism. This approach views a businesswoman’s ‘belief’, ‘habit’ and ‘doubt’ as critical for researching gender related issues in entrepreneurship. This study explains the growth aspects of women-owned manufacturing and services SMEs of Bangladesh with the aim of addressing two neglected research issues. One is the scarcity of studies on growth oriented women entrepreneurs in developing countries. The other is the lack of focus on very successful high-growth women-owned firms in the context of a strong male-dominated economy. This study adopts a framework developed out of the Diana International Project to evaluate the factors influencing the growth of these successful, growing, Bangladeshi women-owned businesses. In order to evaluate the growth process itself, this framework was then modified with growth resources and actions as explained by Edith Penrose in her 1959 seminal book The Theory of Growth of the Firm. This allows for the investigation of the effects of managerial and entrepreneurial abilities in growth, and the identification of how firms achieve growth. A multiple-case design is adopted, covering sixteen successful growth-oriented firms in the manufacturing and services sector. SMEs were studied as the basis for firm growth from initial venture creation, while the sector concentration on manufacturing and services reflects the urban nature of the study in examining firms that exist in the capital city of Dhaka. Data from in-depth interviews and supporting documents were used for the case studies and integrated with the theoretical framework. Themes were categorised and patterns compared against the framework. The results of this research suggest that SME growth is a process which is gradual and iterative, comprising a series of growth strategies and approaches. The framework identifies interactive connection between different growth variables and highlights how industry sector and the national context of a growing economy facilitate growth of women-owned SMEs. The case study based research seeks to advance scholarship in relation to women’s entrepreneurship globally and contribute to the understanding of growth oriented women’s entrepreneurship. Building upon existing knowledge, this research endeavours to generate new insights and advance theoretical discourse by providing richness and subtlety to the knowledge of growth process and opening up new avenues for future research.
