During high-intensity exercise, intracellular creatine phosphate (PCr) is rapidly broken down to maintain adenosine triphosphate turnover. This has lead to the widespread use of creatine monohydrate as a nutritional ergogenic aid. However, the increase in intracellular PCr and the concomitant increase in intracellular water have not been investigated with regard to their effect on active range of movement (ROM). Forty male subjects (age, 24+/-3.2 years) underwent restricted randomization into 2 equal groups, either an intervention group (CS) or a control group (C). The CS group ingested 25 g.day(-1) of creatine monohydrate for 5 days, followed by 5 g.day(-1) for a further 3 days. Before (24 h before starting supplementation (PRE) and after (on the 8th day of supplementation (POST)) this loading phase, both groups underwent goniometry measurement of the shoulder, elbow, hip, and ankle. Data indicated significant reductions in active ROM in 3 movements: shoulder extension (57+/-11.3 degrees PRE vs. 48+/-11.2 degrees POST, p<0.01), shoulder abduction (183.4+/-6.8 degrees PRE vs. 180.3+/-5.1 degrees POST, p<0.05), and ankle dorsiflexion (14.2+/-4.7 degrees PRE vs. 12.1+/-6.4 degrees POST, p<0.01). There was also a significant increase in body mass for the CS group (83.6+/-6.2 kg vs. 85.2+/-6.3 kg, p<0.05). The results suggest that short-term supplementation with creatine monohydrate reduces the active ROM of shoulder extension and abduction and of ankle dorsiflexion. Although the mechanism for this is not fully understood, it may be related to the asymmetrical distribution of muscle mass around those joints.

This task can be very time consuming, particularly when the diatom valves are not abundant. This research was conducted to determine the minimum number of valves to be identified in the diatom assemblages from Rawapening Lake, Central Java, Indonesia. Based on the 314 samples obtained from Rawapening Lake, the diatom efficiency rose above 0.85 upon the minimum count of 300 valves. The number of diatom species identified remained stable after the minimum of 300 valves. Therefore, the minimum number of diatom's valves identified to represent the assemblage for paleolimnological analysis was 300.

It is well known that the mutations in BRCA1 or BRCA2 gene can cause the hereditary breast cancer. However, it is a tedious and expensive task to identify the mutant genes that impact breast cancer due to the large number of genes and very small number of samples. Furthermore, the expressive energy of the subset of genes in comparison to that of one individual gene at a time is considered to have a profound influence in case of breast cancer. In this paper 7 tumors with BRCA1 mutation and 8 tumors with BRCA2 mutation have been used to identify the subset of discriminative genes. A combination of a non-parametric supervised and an unsupervised statistical method is introduced to analyze the gene expressions and the distinctive genes among the highly expressed genes are identified. The most important genes are filtered using the area under the curve (AUC) measure. These filtered genes are then used to build a hidden Markov model (HMM) to analyse their inter-relationship and identify the best subset among them. In addition, Protein-Protein interaction network is generated to analyse the pathways of the identified genes and their link with BRCA1 or BRCA2. Transcription Factors are identified and Gene Set Enrichment Analysis (GSEA) is calculated for the identified genes subset and the results are compared with the results mentioned in other cancer literature. Experimental results suggest that only 8 genes have been identified out of 3226 genes by the proposed hybrid method. Out of the 8 identified genes, 5 have been linked with breast cancer by other studies. Moreover, 7 genes have been associated with numerous diseases that may result in breast cancer. Furthermore, 8 transcription factors were identified that cover the identified genes and BRCA1 and BRCA2. Lastly, GSEA enrichment score of 0.52 is calculated for the identified genes and it is comparatively better considering the small subset of identified genes.

Background. Antibody titres and vaccine effectiveness decline within 6 months after influenza vaccination in older adults. Biannual vaccination may be necessary to provide year-round protection in the tropics, where influenza circulates throughout the year. Methods. Tropical Influenza Control Strategies (TROPICS1) was a single-center, 1:1 randomized, observer-blinded, active-comparator- controlled, superiority study in 200 community-resident adults aged ≥65 years. Participants received a standard-dose trivalent inactivated influenza vaccination (IIV3) at enrollment, and either tetanus-diphtheria-pertussis vaccination or IIV3 6 months later. The primary outcome was the proportion of participants with haemagglutination-inhibition (HI) geometric mean titre (GMT) ≥1:40 1 month after the second vaccination (month 7). Secondary outcomes included GMTs to month 12, the incidence of influenza- like illness (ILI), and adverse reactions after vaccination. Results. At month 7, the proportion of participants with an HI tire ≥1:40 against A/H1N1 increased by 21.4% (95% confidence interval [CI] 8.6-33.4) in the semiannual vaccination group. This proportion was not significantly higher for A/H3N2 (4.3, 95% CI -1.1-10.8) or B (2.1, 95% CI -2.0-7.3). Semiannual vaccination significantly increased GMTs against A/H1N1 and A/H3N2, but not B, at month 7. Participants receiving a repeat vaccination of IIV3 reported a significantly lower incidence of ILI in the 6 months after the second vaccination (relative vaccine effectiveness 57.1%, 95% CI 0.6-81.5). The frequency of adverse events was similar after the first and second influenza vaccinations. Conclusions. Semiannual influenza vaccination in older residents of tropical countries has the potential to improve serological measures of protection against infection. Alternative vaccination strategies should also be studied.

Haemonchus contortus is the most pathogenic nematode of small ruminants. Infection in sheep and goats results in anaemia that decreases animal productivity and can ultimately cause death. The involvement of ruminant-specific galectin-11 (LGALS-11) and galectin-14 (LGALS-14) has been postulated to play important roles in protective immune responses against parasitic infection; however, their ligands are unknown. In the current study, LGALS-11 and LGALS-14 ligands in H. contortus were identified from larval (L4) and adult parasitic stages extracts using immobilised LGALS-11 and LGALS-14 affinity column chromatography and mass spectrometry. Both LGALS-11 and LGALS-14 bound more putative protein targets in the adult stage of H. contortus (43 proteins) when compared to the larval stage (two proteins). Of the 43 proteins identified in the adult stage, 34 and 35 proteins were bound by LGALS-11 and LGALS-14, respectively, with 26 proteins binding to both galectins. Interestingly, hematophagous stage-specific sperm-coating protein and zinc metalloprotease (M13), which are known vaccine candidates, were identified as putative ligands of both LGALS-11 and LGALS- 14. The identification of glycoproteins of H. contortus by LGALS-11 and LGALS-14 provide new insights into host-parasite interactions and the potential for developing new interventions.

Genetic network reverse engineering has been an area of intensive research within the systems biology community during the last decade. With many techniques currently available, the task of validating them and choosing the best one for a certain problem is a complex issue. Current practice has been to validate an approach on in-silico synthetic data sets, and, wherever possible, on real data sets with known ground-truth. In this study, we highlight a major issue that the validation of reverse engineering algorithms on small benchmark networks very often results in networks which are not statistically better than a randomly picked network. Another important issue highlighted is that with short time series, a small variation in the pre-processing procedure might yield large differences in the inferred networks. To demonstrate these issues, we have selected as our case study the IRMA in-vivo synthetic yeast network recently published in Cell. Using Fisher's exact test, we show that many results reported in the literature on reverse-engineering this network are not significantly better than random. The discussion is further extended to some other networks commonly used for validation purposes in the literature. The results presented in this study emphasize that studies carried out using small genetic networks are likely to be trivial, making it imperative that larger real networks be used for validating and benchmarking purposes. If smaller networks are considered, then the results should be interpreted carefully to avoid over confidence. Paleolimnological techniques can reveal long-term perturbations and associated stable state transitions of lake ecosystems. However, such transitions are difficult to predict since changes to lake ecosystems can be abrupt or gradual. This study examined whether there were past transitions in the ecological regime of Kings Billabong, a shallow River Murray wetland in southeast Australia. A 94-cm-long core, covering c. 90 years of age, was analysed at 1 cm resolution for subfossil cladocerans, diatoms and other proxies. Prior to river regulation (c. 1930), the littoral to planktonic ratios of cladocerans and diatoms, and bulk sediment delta C-13 values were high, while the period from c. 1930 to c. 1970 experienced considerable changes to the wetland ecosystem. The abrupt nature of changes of planktonic cladocerans and diatoms, particularly after the onset of river regulation (1930s), was triggered by inundation, high rates of sedimentation and shifts in bulk sediment delta N-15 values. However, the transition of a once littoral-dominated community, to one favouring an increasingly turbid, plankton-dominated trophic condition following river regulation was relatively slow and lasted for decades. The progression to a new regime was likely delayed by the partial recovery of submerged plant communities and related internal dynamics.
